The arginine residue is moderately conserved and there is a moderate physicochemical difference between arginine and tryptophan. This variant is present in population databases (rs775651175, gnomAD 0.006%). This missense change has been observed in individual(s) with breast cancer (PMID: 28076423). ClinVar contains an entry for this variant (Variation ID: 407019). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt RECQL4 protein function with a positive predictive value of 80%.
