This protein is an important cell cycle checkpoint kinase that phosphorylates; thus, it functions as a regulator of a wide variety of downstream proteins, including tumor suppressor proteins p53 and BRCA1, checkpoint kinase CHK2, checkpoint proteins RAD17 and RAD9, and DNA repair protein NBS1. This protein and the closely related kinase ATR are thought to be master controllers of cell cycle checkpoint signaling pathways that are required for cell response to DNA damage and for genome stability. Mutations in this gene are associated with ataxia telangiectasia, an autosomal recessive disorder. [provided by RefSeq, Aug 2010]

The valine at codon 2439 is replaced by alanine, an amino acid with similar properties. This alteration has been detected in 2/4112 breast cancer patients and 0/2399 healthy control individuals across numerous studies (Tavtigian SV et al. Am J Hum Genet. 2009 Oct; 85(4):427-46). This alteration was also identified in 0/87 patients with CS or BRRS and 1/3476 patients from The Cancer Genome Atlas (TCGA) (Yehia L et al. PLoS Genet, 2018 04;14:e1007352) as well as in 1/104 cases of familial cancer patients in an Italian cohort (Paduano F et al. Genes (Basel) 2022 Jul;13(7)). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.7316T>C has been reported in the literature in individuals affected with Breast Cancer (Thorstenson_2003, Akcay_2020, Bandeira_2020) as well as one individual with severe short stature of unknown etiology (Guo_2014). These reports do not provide unequivocal conclusions about association of the variant with Breast Cancer. One co-occurrence with another pathogenic variant has been reported (BRCA2 c.5909C>A, p.Ser1970X, Bandeira_2020), providing supporting evidence for a benign role.
