Variant summary
Our verdict is Uncertain significance. The variant received 4 ACMG points: 5P and 1B. PM1PM2PP2BP4
The NM_000492.4(CFTR):c.4142A>C(p.Tyr1381Ser) variant causes a missense change. The variant allele was found at a frequency of 0.00000139 in 1,437,792 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a benign outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. Y1381C) has been classified as Uncertain significance.

Cystic fibrosis Uncertain:1
The p.Y1381S variant (also known as c.4142A>C), located in coding exon 26 of the CFTR gene, results from an A to C substitution at nucleotide position 4142. The tyrosine at codon 1381 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
