The p.Asp334Asn var iant in TMPRSS3 has not been previously reported in individuals with hearing los s, but was identified in 1/66272 European chromosomes by the Exome Aggregation C onsortium (ExAC, http://exac.broadinstitute.org). Aspartic acid (Asp) at positio n 334 is conserved in most mammals, but not evolutionarily distant species, and elephants carry an asparagine (Asn) at this position, suggesting that this chang e may be tolerated. Additional computational prediction tools do not provide str ong support for or against an impact to the protein. In summary, while the clini cal significance of the p.Asp334Asn variant is uncertain, the amino acid conserv ation data suggests that it is more likely to be benign. -
