not specified Uncertain:1
The c.211G>C (p.G71R) alteration is located in exon 2 (coding exon 1) of the FLRT1 gene. This alteration results from a G to C substitution at nucleotide position 211, causing the glycine (G) at amino acid position 71 to be replaced by an arginine (R). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Peripheral neuropathy Uncertain:1
In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). This variant has not been reported in the literature in individuals with FLRT1-related disease. This variant is present in population databases (rs777177875, ExAC 0.05%). This sequence change replaces glycine with arginine at codon 71 of the FLRT1 protein (p.Gly71Arg). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and arginine. -
