This sequence change replaces histidine with arginine at codon 804 of the BRIP1 protein (p.His804Arg). The histidine residue is highly conserved and there is a small physicochemical difference between histidine and arginine. This variant is present in population databases (rs777277034, ExAC 0.006%). This variant has not been reported in the literature in individuals affected with BRIP1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The p.H804R variant (also known as c.2411A>G), located in coding exon 16 of the BRIP1 gene, results from an A to G substitution at nucleotide position 2411. The histidine at codon 804 is replaced by arginine, an amino acid with highly similar properties. This alteration was observed within individuals with a personal history of breast cancer. (Easton DF et al. J Med Genet, 2016 May;53:298-309). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
