This alteration results from a G to T substitution at nucleotide position 983, causing the serine (S) at amino acid position 328 to be replaced by an isoleucine (I). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.

The S328I variant in the AHI1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The S328I variant is observed in 26/30780 (0.084%) alleles from individuals of South Asian background, in large population cohorts (Lek et al., 2016). The S328I variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is not conserved. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret S328I as a variant of uncertain significance.
