The p.Arg652Gln variant in POLD1 has not been previously reported in individuals with colorectal cancer, but has been identified in 1/10178 of African chromosom es by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org; db SNP rs778037523). In vitro functional studies in a rat hepatoma cell line that c arried this change at the position corresponding to human amino acid 652 provide some evidence that the p.Arg652Gln variant may impact protein function (Popanda 1999). However, these types of assays may not accurately represent biological f unction. Computational prediction tools and conservation analysis do not provide strong support for or against an impact to the protein. In summary, the clinica l significance of the p.Arg652Gln variant is uncertain. -
Colorectal cancer, susceptibility to, 10 Uncertain:1
This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 652 of the POLD1 protein (p.Arg652Gln). This variant is present in population databases (rs778037523, gnomAD 0.008%). This variant has not been reported in the literature in individuals affected with POLD1-related conditions. ClinVar contains an entry for this variant (Variation ID: 239262). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt POLD1 protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The p.R652Q variant (also known as c.1955G>A), located in coding exon 15 of the POLD1 gene, results from a G to A substitution at nucleotide position 1955. The arginine at codon 652 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
