Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251452 control chromosomes (gnomAD). c.1169G>C has been reported in the literature in homozygous and compound heterozygous individuals affected with Glutaric Acidemia Type 1 (examples: Spenger_2021, Gurbuz_2020, Kurkina_2020, Wang_2014). At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in <10% of normal activity in e Coli (example: Goodman_1998). Different variant affecting this residue (c.1168G>C, p.Gly390Arg) has been classified pathogenic in ClinVar (CV ID193799). These data indicate that the variant is very likely to be associated with disease. The following publications have been ascertained in the context of this evaluation (PMID: 9711871, 32777384, 34207159, 32240488, 24332224).
