The p.K241T variant (also known as c.722A>C), located in coding exon 9 of the MLH1 gene, results from an A to C substitution at nucleotide position 722. The lysine at codon 241 is replaced by threonine, an amino acid with similar properties. This variant has been identified in a cohort of 8085 Chinese breast cancer patients (Hu L et al. NPJ Breast Cancer, 2022 Apr;8:52). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -
