This variant has not been reported in the literature in individuals affected with SPG11-related conditions. ClinVar contains an entry for this variant (Variation ID: 423138). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. -

The c.5772_5781dup10 pathogenic variant in the SPG11 gene causes a frameshift starting with codon Histidine 1928, changes this amino acid to a Glycine residue and creates a premature Stop codon at position 15 of the new reading frame, denoted p.H1928GfsX15. This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. This variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). Therefore, we interpret c.5772_5781dup10 as a pathogenic variant. -
