The c.10685G>A (p.R3562H) alteration is located in exon 76 (coding exon 76) of the RYR3 gene. This alteration results from a G to A substitution at nucleotide position 10685, causing the arginine (R) at amino acid position 3562 to be replaced by a histidine (H). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces arginine with histidine at codon 3562 of the RYR3 protein (p.Arg3562His). The arginine residue is moderately conserved and there is a small physicochemical difference between arginine and histidine. This variant is present in population databases (rs780080294, ExAC 0.01%). This variant has not been reported in the literature in individuals with RYR3-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
