Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign. The p.T449M variant (also known as c.1346C>T), located in coding exon 13 of the POLE gene, results from a C to T substitution at nucleotide position 1346. The threonine at codon 449 is replaced by methionine, an amino acid with similar properties. This alteration was detected in an individual with a personal and family history of colorectal cancer who underwent whole-exome sequencing (Xu P et al. JCI Insight, 2021 09;6). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
