This variant is reported in 0.0099% of alleles in individuals of Ashkenazi Jewish descent in gnomAD (http://gnomad.broadinstitute.org/variant/2-152474966-C-T). Nonsense variants in NEB are expected to be pathogenic. This variant is interpreted as pathogenic. -
Nemaline myopathy Pathogenic:1
The p.Trp3633Ter variant in NEB has not been previously reported in the literature in individuals with nemaline myopathy, but has been identified in 0.007% (2/29604) of Ashkenazi Jewish chromosomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadinstitute.org; dbSNP ID: rs780343350). Although this variant has been seen in the general population in a heterozygous state, its frequency is low enough to be consistent with a recessive carrier frequency. This variant has also been reported in ClinVar (Variation ID: 581881) and has been interpreted as pathogenic by Invitae. This nonsense variant leads to a premature termination codon at position 3633, which is predicted to lead to a truncated or absent protein. Loss of function of the NEB gene is an established disease mechanism in autosomal recessive nemaline myopathy. In summary, although additional studies are required to fully establish its clinical significance, this variant is likely pathogenic for autosomal recessive nemaline myopathy.
