The c.394C>T (p.R132C) alteration is located in exon 7 (coding exon 7) of the CD247 gene. This alteration results from a C to T substitution at nucleotide position 394, causing the arginine (R) at amino acid position 132 to be replaced by a cysteine (C). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces arginine with cysteine at codon 132 of the CD247 protein (p.Arg132Cys). The arginine residue is highly conserved and there is a large physicochemical difference between arginine and cysteine. This variant is present in population databases (rs781510519, ExAC 0.02%). This variant has not been reported in the literature in individuals affected with CD247-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
