The variant was identified in dbSNP (ID: rs781777438), ClinVar (uncertain significance by Invitae with associated phenotype of Werner Syndrome), and in LOVD 3.0. The variant was also identified in control databases in 7 of 279294 chromosomes at a frequency of 0.000025 (Genome Aggregation Database Feb 27, 2017). The variant was observed in the following populations: European (non-Finnish) in 6 of 127798 chromosomes (freq: 0.000047) and South Asian in 1 of 30268 chromosomes (freq: 0.000033); it was not observed in the African, Latino, Ashkenazi Jewish, East Asian, European (Finnish) and Other populations. This variant is an in-frame deletion resulting in the removal of a glutamic acid (glu) residue at codons 509 and 510 in a repeat region; the impact of this alteration on WRN protein function is not known. The variant occurs outside of the splicing consensus sequence and in silico programs (SpliceSiteFinder, MaxEntScan, NNSPLICE, and GeneSplicer) do not predict a difference in splicing. In summary, based on the above information the clinical significance of this variant cannot be determined with certainty at this time although we would lean towards a more benign role for this variant.
