The c.9522_9523delTGinsAT pathogenic mutation, located in coding exon 25 of the BRCA2 gene, results from an in-frame deletion of TG and insertion of AT at nucleotide positions 9522 to 9523. This change results in a termination codon at amino acid position 3175 within coding exon 10 (p.N3174_E3175delinsK*). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediatedmRNAdecay.As such, this alteration is interpreted as a disease-causing mutation. -
