This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 656 of the RAD50 protein (p.Arg656Gln). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with RAD50-related conditions. ClinVar contains an entry for this variant (Variation ID: 186977). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. The p.R656Q variant (also known as c.1967G>A), located in coding exon 12 of the RAD50 gene, results from a G to A substitution at nucleotide position 1967. The arginine at codon 656 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species.
