The c.1921_1923delGAA variant (also known as p.E641del) is located in coding exon 4 of the MSH6 gene. This variant results from an in-frame GAA deletion at nucleotide positions 1921 to 1923. This results in the in-frame deletion of a glutamic acid at codon 641. This variant was reported in one Puerto Rican patient with breast cancer who previously tested negative for BRCA1/2 mutations (Dutil J et al. Sci Rep, 2019 11;9:17769). The deleted amino acid position is poorly conserved in available vertebrate species. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
