Mutations in this gene are associated with a variety of human cancers, including hereditary cancers such as Li-Fraumeni syndrome. Alternative splicing of this gene and the use of alternate promoters result in multiple transcript variants and isoforms. The aspartic acid at codon 259 is replaced by glycine, an amino acid with similar properties. This alteration has been reported in the literature in a cohort of individuals affected with chronic lymphocytic leukemia (Pekova S et al. Leuk Res, 2011 Jul;35:889-98). Functional studies assessing the transactivation capacity of the alteration have reported the variant as functional (Kotler E et al. Mol Cell, 2018 07;71:178-190.e8, Giacomelli AO et al. Nat Genet, 2018 10;50:1381-1387) or partially functional (Kato S et al. Proc Natl Acad Sci U S A, 2003 Jul;100:8424-9). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
