This sequence change replaces methionine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 326 of the ACADM protein (p.Met326Thr). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with medium-chain acyl CoA dehydrogenase deficiency (PMID: 8198141, 23028790). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is also known as M301T. ClinVar contains an entry for this variant (Variation ID: 189019). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ACADM protein function with a positive predictive value of 80%. Experimental studies have shown that this missense change affects ACADM function (PMID: 8198141, 8535441). This variant disrupts the p.Met326 amino acid residue in ACADM. Other variant(s) that disrupt this residue have been observed in individuals with ACADM-related conditions (PMID: 32793418), which suggests that this may be a clinically significant amino acid residue.
