This changes the amino acid from a tryptophan to a stop codon within coding exon 20. This alteration has been identified in multiple individuals with cystic fibrosis (Shoshani T et al. Hum. Mol. Genet., 1994 Apr;3:657-8; Martínez-Hernández A et al. BMC Med Genomics, 2019 05;12:68). This alteration is also associated with pancreatic insufficiency and elevated sweat chloride levels (Sosnay PR et al. Nat. Genet., 2013 Oct;45:1160-7) and with congenital absence of the vas deferens (Saldaña-Alvarez Y et al. Genet Test Mol Biomarkers, 2012 Apr;16:292-6). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay.
