Multiple endocrine neoplasia, type 1 Pathogenic:1
This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 12 of the MEN1 protein (p.Pro12Leu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with multiple endocrine neoplasia type 1 (PMID: 9215689, 31658439; internal data). ClinVar contains an entry for this variant (Variation ID: 200969). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt MEN1 protein function with a positive predictive value of 95%. Experimental studies have shown that this missense change affects MEN1 function (PMID: 12509449, 15254225, 21264250, 21819486, 22090276). For these reasons, this variant has been classified as Pathogenic. -
