Variant summary: ACADVL c.535G>A (p.Gly179Arg) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251462 control chromosomes (gnomAD). c.535G>A has been reported in the literature in newborn screening cases affected with Very Long Chain Acyl-CoA Dehydrogenase Deficiency (D'Annibale_2022). These data indicate that the variant may be associated with disease. This publication also reports experimental evidence evaluating an impact on protein function, finding that the variant results in <10% of normal enzymatic activity. The following publication has been ascertained in the context of this evaluation (PMID: 35218577). ClinVar contains an entry for this variant (Variation ID: 839572).
