Genes affected
OCA2 (HGNC:8101): (OCA2 melanosomal transmembrane protein) This gene encodes the human homolog of the mouse p (pink-eyed dilution) gene. The encoded protein is believed to be an integral membrane protein involved in small molecule transport, specifically tyrosine, which is a precursor to melanin synthesis. It is involved in mammalian pigmentation, where it may control skin color variation and act as a determinant of brown or blue eye color. Mutations in this gene result in type 2 oculocutaneous albinism. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jul 2014]

Pathogenic, criteria provided, single submitterclinical testingInvitaeJan 15, 2024This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 782 of the OCA2 protein (p.Gly782Arg). This variant is present in population databases (rs797045839, gnomAD 0.008%). This missense change has been observed in individual(s) with oculocutaneous albinism (PMID: 19865097, 31077556, 34838614). ClinVar contains an entry for this variant (Variation ID: 211770). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt OCA2 protein function with a positive predictive value of 80%. For these reasons, this variant has been classified as Pathogenic. -
