Variant summary: BRCA1 c.2935C>T (p.Arg979Cys) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 2.4e-05 in 251168 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2935C>T has been reported in the literature in individuals affected with breast/ovarian cancer (e.g. Anczukow_2008, Judkins_2005) and has recently been classified as IARC class 1 (Benign) based upon the French national COVAR (cosegregation variant) study that utilized a multifactorial model combining different associations between VUSs and cancer, including cosegregation data (Caputo_2021). These report(s) do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. At-least one co-occurrence with another pathogenic variant(s) have been reported in the UMD database (BRCA2 c.771_775delTCAAA, p.Asn257LysfsX17), providing supporting evidence for a benign role. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 16267036, 16518693, 24728327, 23704879, 23893897, 18273839, 31112341, 31294896, 34749799, 34597585).
