The serine at codon 628 is replaced by arginine, an amino acid with dissimilar properties. This alteration has been classified as likely to be neutral or of little clinical significance based on a classification system using interspecific sequence variation (Abkevich V et al. J. Med. Genet., 2004 Jul;41:492-507). This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
