The lysine at codon 345 is replaced by glutamic acid, an amino acid with similar properties. In a population-based sample of young women with contralateral breast cancer and unilateral breast cancer, this alteration was detected in 1/1398 women with unilateral breast cancer but not in 705 women with contralateral breast cancer (Borg A et al. Hum Mutat. 2010 Mar;31(3):E1200-40). Additionally, this alteration was observed with an allele frequency of 0.00014 in 7,051 unselected female breast cancer patients and was observed with an allele frequency of 0.0000 in 11,241 female controls of Japanese ancestry (Momozawa Y et al. Nat Commun, 2018 10;9:4083). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis.
