The p.E471K variant (also known as c.1411G>A), located in coding exon 9 of the BRCA2 gene, results from a G to A substitution at nucleotide position 1411. The glutamic acid at codon 471 is replaced by lysine, an amino acid with similar properties. This alteration was identified to co-occur with a pathogenic BRCA1 alteration in 1 of 2168 Turkish individuals undergoing BRCA1/2 genetic testing based on a personal and/or family history of HBOC-related cancer (Bisgin A et al. Breast, 2022 Oct;65:15-22). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This variant has not been reported in the literature in individuals with BRCA2-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glutamic acid with lysine at codon 471 of the BRCA2 protein (p.Glu471Lys). The glutamic acid residue is weakly conserved and there is a small physicochemical difference between glutamic acid and lysine. -
