The alanine at codon 495 is replaced by threonine, an amino acid with similar properties. In one study, this variant was detected in a 53 year-old Argentinian proband with breast cancer prior to 50 years of age, whose family history included four sisters with breast cancer prior to 50 years of age (Giannini G et al. Breast Cancer Res. Treat. 2006; 100:83-91). Additional studies have detected this variant in hereditary breast and/or ovarian cancer families from Italian and Brazilian cohorts (Silva FC et al. BMC Med. Genet., 2014 May;15:55; Capalbo C et al. Ann. Oncol., 2006 Jun;17 Suppl 7:vii34-40). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
