The p.S196N variant (also known as c.587G>A), located in coding exon 6 of the BRCA2 gene, results from a G to A substitution at nucleotide position 587. The serine at codon 196 is replaced by asparagine, an amino acid with highly similar properties. This alteration was shown to cause slightly increased exon skipping (25% and 28%) compared to wild type in splicing mini gene assays (Gaildrat P. J. Med. Genet.. 2012 Oct;49(10):609-17; Di Giacomo D et al. Hum Mutat, 2013 Nov;34:1547-57). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
This missense variant replaces serine with asparagine at codon 196 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). Functional studies have shown that this variant causes a slight increase in exon 7 skipping in mini-gene assays (PMID: 22962691, 23983145). However, clinical relevance of this observation is not known. This variant was reported in an individual affected with breast cancer (PMID: 22962691). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

This sequence change replaces serine, which is neutral and polar, with asparagine, which is neutral and polar, at codon 196 of the BRCA2 protein (p.Ser196Asn). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with breast cancer (PMID: 22962691). ClinVar contains an entry for this variant (Variation ID: 51958). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRCA2 protein function with a negative predictive value of 95%. Experimental studies have shown that this missense change does not substantially affect BRCA2 function (PMID: 32286328). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
