Genes affected
BRCA2 (HGNC:1101): (BRCA2 DNA repair associated) Inherited mutations in BRCA1 and this gene, BRCA2, confer increased lifetime risk of developing breast or ovarian cancer. Both BRCA1 and BRCA2 are involved in maintenance of genome stability, specifically the homologous recombination pathway for double-strand DNA repair. The largest exon in both genes is exon 11, which harbors the most important and frequent mutations in breast cancer patients. The BRCA2 gene was found on chromosome 13q12.3 in human. The BRCA2 protein contains several copies of a 70 aa motif called the BRC motif, and these motifs mediate binding to the RAD51 recombinase which functions in DNA repair. BRCA2 is considered a tumor suppressor gene, as tumors with BRCA2 mutations generally exhibit loss of heterozygosity (LOH) of the wild-type allele. [provided by RefSeq, May 2020]

Uncertain significance, criteria provided, single submitterclinical testingAmbry GeneticsJul 05, 2023The p.V2205M variant (also known as c.6613G>A), located in coding exon 10 of the BRCA2 gene, results from a G to A substitution at nucleotide position 6613. The valine at codon 2205 is replaced by methionine, an amino acid with highly similar properties. This alteration has been reported in multiple individuals with a personal history of breast and/or ovarian cancer (Borg A et al. Hum. Mutat. 2010 Mar;31:E1200-40; Capanu M et al. Genet. Epidemiol., 2011 Jul;35:389-97; Cunningham JM et al. Sci Rep. 2014 Feb;4:4026; Ruiz A et al. Biomed Res Int. 2014 Jun;2014:54254; Minucci A et al. Expert Rev. Mol. Diagn. 2015 Aug;15:1383-4031) and in a cohort of individuals referred for genetic analysis of the BRCA1 and BRCA2 genes (Apessos A et al. Cancer Genet, 2018 Jan;220:1-12). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

Uncertain significance, criteria provided, single submitterclinical testingWomen's Health and Genetics/Laboratory Corporation of America, LabCorpMar 03, 2017Variant summary: The BRCA2 c.6613G>A (p.Val2205Met) variant causes a missense change involving the alteration of a non-conserved nucleotide, which 4/5 in silico tools predict a benign outcome, although these predictions have yet to be functionally assessed. The variant of interest was observed in the large, broad control population, ExAC, with an allele frequency of 5/120326 (1/24038), which does not exceed the estimated maximal expected allele frequency for a pathogenic BRCA2 variant of 1/1333. Multiple publications have cited the variant in affected individuals (BrC, OvC, HBOC), however, with limited information (ie lack of co-occurrence and cosegregation data). In addition, multiple clinical diagnostic laboratories/reputable databases classified this variant as uncertain significance. Therefore, until additional information becomes available (ie, clinical and functional studies), the variant of interest has been classified as a "Variant of Uncertain Significance (VUS)." -

Uncertain significance, criteria provided, single submitterclinical testingQuest Diagnostics Nichols Institute San Juan CapistranoMay 11, 2023The frequency of this variant in the general population, 0.000054 (6/111976 chromosomes, http://gnomad.broadinstitute.org), is uninformative in assessment of its pathogenicity. In the published literature, the variant has been reported in individuals with breast cancer (PMID: 20104584 (2010), 21520273 (2011), 29335924 (2018)), ovarian cancer (PMID: 24504028 (2014), 26306726 (2015)), and familial breast and/or ovarian cancer (PMID: 25136594 (2014), 29310832 (2018), 31159747 (2019)). Additionally, a multifactorial likelihood analysis predicts that the variant is likely benign (PMID: 31131967 (2019)). Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign. Based on the available information, we are unable to determine the clinical significance of this variant. -
