Hereditary cancer-predisposing syndrome Uncertain:2Benign:1
Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Aug 29, 2023 | The p.N2266H variant (also known as c.6796A>C), located in coding exon 10 of the BRCA2 gene, results from an A to C substitution at nucleotide position 6796. The asparagine at codon 2266 is replaced by histidine, an amino acid with similar properties. This alteration was reported in a cohort of 402 hereditary breast and ovarian cancer patients (Schenkel LC et al. J Mol. Diagn. 2016 09;18:657-667). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
Uncertain significance, criteria provided, single submitter | clinical testing | Color Diagnostics, LLC DBA Color Health | Aug 14, 2023 | This missense variant replaces asparagine with histidine at codon 2266 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been performed for this variant. This variant has been reported in an individual with personal or family history of breast and/or ovarian cancer (PMID: 27376475) and reported in three times in the Canadian Open Genetics Repository (PMID: 28726806). This variant has also been described in a multifactorial analysis with low prior probability of pathogenicity based on co-occurrence and family history likelihood ratios, however, it did not provide a definitive classification nor a posterior probability (PMID: 31131967). This variant has been identified in 1/250702 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. - |

BRCA2-related cancer predisposition Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | All of Us Research Program, National Institutes of Health | Jul 29, 2024 | This missense variant replaces asparagine with histidine at codon 2266 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). Splice site prediction tools suggest that this variant may not impact RNA splicing. The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. - |
not provided Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | GeneDx | Sep 03, 2019 | Not observed at a significant frequency in large population cohorts (Lek 2016); In silico analysis, which includes protein predictors and evolutionary conservation, supports that this variant does not alter protein structure/function; Observed in individuals with a personal or family history of breast and/or ovarian cancer (Schenkel 2016); This variant is associated with the following publications: (PMID: 27376475, 25348012) - |
Malignant tumor of breast Uncertain:1
Uncertain significance, no assertion criteria provided | clinical testing | Department of Pathology and Laboratory Medicine, Sinai Health System | - | The p.Asn2266His variant has not been reported in the literature, but has been recorded in the UMD (x1) and BIC (x2) databases. The variant is listed in the dbSNP database (ID#:rs80358905) as coming from a "clinical source" but no frequency information was provided, and so the prevalence of this variant in the population is not known. This residue is conserved in mammals; however, computational analyses (PolyPhen, SIFT, AlignGVGD, BLOSUM) provide inconsistent predictions regarding the impact to the protein and this information is not very predictive of pathogenicity. In summary, based on the above information, the clinical significance of this variant cannot be determined with certainty at this time. This variant is classified as a variant of unknown significance. - |
Hereditary breast ovarian cancer syndrome Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Nov 10, 2023 | This sequence change replaces asparagine, which is neutral and polar, with histidine, which is basic and polar, at codon 2266 of the BRCA2 protein (p.Asn2266His). This variant is present in population databases (rs80358905, gnomAD 0.0009%). This missense change has been observed in individual(s) with hereditary breast and ovarian cancer (HBOC) (PMID: 10923033, 27376475). ClinVar contains an entry for this variant (Variation ID: 52184). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRCA2 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. - |
