The p.N2266H variant (also known as c.6796A>C), located in coding exon 10 of the BRCA2 gene, results from an A to C substitution at nucleotide position 6796. The asparagine at codon 2266 is replaced by histidine, an amino acid with similar properties. This alteration was reported in a cohort of 402 hereditary breast and ovarian cancer patients (Schenkel LC et al. J Mol. Diagn. 2016 09;18:657-667). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This variant has also been described in a multifactorial analysis with low prior probability of pathogenicity based on co-occurrence and family history likelihood ratios, however, it did not provide a definitive classification nor a posterior probability (PMID: 31131967). This variant has been identified in 1/250702 chromosomes in the general population by the Genome Aggregation Database (gnomAD).
