The p.S3014* pathogenic mutation (also known as c.9041C>A), located in coding exon 22 of the BRCA2 gene, results from a C to A substitution at nucleotide position 9041. This changes the amino acid from a serine to a stop codon within coding exon 22. This alteration was identified amongst a cohort of 3984 Chinese women diagnosed with a breast cancer undergoing BRCA1/2 genetic testing (Yao L et al. J Hum Genet, 2022 Nov;67:639-642). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
