This deletion of two nucleotides in BRCA2 is denoted c.1496_1497delAG at the cDNA level and p.Gln499ArgfsX14 (Q499RfsX14) at the protein level. Using alternate nomenclature, this variant would be defined as BRCA2 1724delAG. The normal sequence, with the bases that are deleted in brackets, is TTTC[delAG]GGTA. The deletion causes a frameshift which changes a Glutamine to an Arginine at codon 499, and creates a premature stop codon at position 14 of the new reading frame. This variant is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. BRCA2 c.1496_1497delAG has been reported in a family with male breast cancer (Coppa 2014). We consider this variant to be pathogenic. -
Hereditary cancer-predisposing syndrome Pathogenic:1
The c.1496_1497delAG pathogenic mutation, located in coding exon 9 of the BRCA2 gene, results from a deletion of two nucleotides at nucleotide positions 1496 to 1497, causing a translational frameshift with a predicted alternate stop codon (p.Q499Rfs*14). This mutation, also designated as 1724delAG, has been seen in unrelated breast cancer families, including a case of male breast cancer (Bayraktar S et al. Cancer, 2012 Mar;118:1515-22; Coppa A et al. Breast Cancer Res. Treat., 2014 Dec;148:629-35). This alteration was also identified in multiple families in a large, worldwide study of BRCA1/2 mutation positive families (Rebbeck TR et al. Hum. Mutat., 2018 05;39:593-620). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -

Hereditary breast ovarian cancer syndrome Pathogenic:1
This sequence change creates a premature translational stop signal (p.Gln499Argfs*14) in the BRCA2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with male breast cancer (PMID: 25395318). ClinVar contains an entry for this variant (Variation ID: 51134). For these reasons, this variant has been classified as Pathogenic. -
