This variant has been reported in individuals affected with breast cancer (PMID: 25452441, 26681682, 27153395) or prostate cancer (PMID: 21952622). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of BRCA2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic. -
The c.2836_2837delGA pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a deletion of two nucleotides at nucleotide positions 2836 to 2837, causing a translational frameshift with a predicted alternate stop codon (p.D946Ffs*12). This alteration was identified in individuals diagnosed with prostate cancer and breast cancer (Kote-Jarai, Z et al. Br J Cancer. 2011 Oct 11;105(8):1230-4; Couch Fj et al. J Clin Oncol. 2015 Feb 1;33(4):304-11; Eccles DM et al. Ann Oncol, 2016 Mar;27:467-73). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -

Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant was absent in 250652 control chromosomes. c.2836_2837delGA has been reported in the literature in individuals affected with Breast and Ovarian Cancer (Couch_2015, Eccles_2016) and Prostate Cancer (Kote-Jarai_2011, Castro_2012). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Five clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar (after 2014) and classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -
