This sequence change deletes 4 nucleotides in exon 11 of BRCA2 mRNA (c.3189_3192delGTCA), causing a frameshift after codon 1064 and the creation of a premature translation stop signal 12 amino acid residues later (p.Ser1064Leufs*12). This is expected to result in an absent or disrupted protein product. Truncating variants in BRCA2 are known to be pathogenic. The mutation database ClinVar contains entries for this variant (Variation ID: 51418). This variant is also known as 3417del4 in the literature using alternative nomenclature. -
The BRCA2 c.3189_3192del (p.Ser1064Leufs*12) variant alters the translational reading frame of the BRCA2 mRNA and causes the premature termination of BRCA2 protein synthesis. This variant has been reported in the published literature in individuals with breast cancer (PMID: 35220195 (2022), 34645131 (2022), 31174498 (2019), 29470806 (2018), 29335924 (2018), 26911350 (2016)), esophagus squamous cell carcinoma (PMID: 29625052 (2018)), and male breast cancer (PMID: 10505028 (1999)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Based on the available information, this variant is classified as pathogenic. -

This sequence change creates a premature translational stop signal (p.Ser1064Leufs*12) in the BRCA2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with breast cancer (PMID: 10505028, 26911350). This variant is also known as c.3186_3189delTCAG or 3414delTCAG. ClinVar contains an entry for this variant (Variation ID: 51418). For these reasons, this variant has been classified as Pathogenic. -

The c.3189_3192delGTCA pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a deletion of 4 nucleotides at nucleotide positions 3189 to 3192, causing a translational frameshift with a predicted alternate stop codon (p.S1064Lfs*12). This alteration has been reported in multiple breast and/or ovarian cancer families (Balci A et al. Eur. J. Cancer, 1999 May;35:707-10; Jakimovska M et al. Breast Cancer Res. Treat., 2018 Apr;168:745-753; Rebbeck TR et al. Hum. Mutat., 2018 05;39:593-620; Singh J et al. Breast Cancer Res. Treat., 2018 Jul;170:189-196; Cao WM et al. BMC Cancer, 2019 Jun;19:551; Farra C et al. Hered Cancer Clin Pract, 2019 Jan;17:4). Of note, this alteration is also designated as 3414del4 and c.3186_3189delTCAG in published literature. In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
