The c.3570delG pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a deletion of one nucleotide at nucleotide position 3570, causing a translational frameshift with a predicted alternate stop codon (p.K1191Sfs*6). This alteration has been reported in a Filipino patient with a personal and family history of breast cancer (De Leon Matsuda ML et al. Int. J. Cancer, 2002 Apr;98:596-603). This alteration was also identified in a large, worldwide study of BRCA1/2 mutation positive families (Rebbeck TR et al. Hum. Mutat., 2018 05;39:593-620). This alteration is also reported as 3798delG in the published literature. In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
This variant deletes 1 nucleotide in exon 11 of the BRCA2 gene, creating a frameshift and premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. To our knowledge, functional studies have not been reported for this variant. This variant has been detected in at least one individual each affected with breast or ovarian cancer (PMID: 11920621, 32872764) and in two suspected hereditary breast and ovarian cancer families (PMID: 29446198). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of BRCA2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic. -

Variant summary: BRCA2 c.3570delG (p.Lys1191SerfsX6) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant was absent in 251026 control chromosomes. c.3570delG has been reported in the literature in individuals affected with Hereditary Breast And Ovarian Cancer Syndrome or prostate cancer (De Leon Matsuda_2002, Kwong_2015, Nguyen-Dumont_2020, Rebbeck_2018). These data indicate that the variant is likely to be associated with disease. Six submitters, including an expert panel (ENIGMA), have submitted clinical-significance assessments for this variant to ClinVar after 2014, and classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -
