Variant summary: BRCA2 c.5454delA (p.Cys1820AlafsX20) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. The variant was absent in 251092 control chromosomes (gnomAD). The variant, c.5454delA, has been reported in the literature in individuals with personal- or family history of Hereditary Breast and Ovarian Cancer (Lubinski_2004, Malone_2006). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Three submitters, including one expert panel (ENIGMA), have provided clinical-significance assessments for this variant in ClinVar after 2014 (without evidence for independent evaluation), and classified the variant as pathogenic. This variant has been identified in multiple families with breast and/or ovarian cancer diagnoses (Lubinski J et al. Fam. Cancer. 2004;3:1-10; Malone KE et al. Cancer Res, 2006 Aug;66:8297-308).
