The c.6335_6336delGA pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a deletion of two nucleotides at nucleotide positions 6335 to 6336, causing a translational frameshift with a predicted alternate stop codon (p.R2112Kfs*16). This mutation has been seen in one case report of a patient with ovarian cancer (Rose PG et al. Gynecol Oncol, 2000 May;77:319-20). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
