To our knowledge, this variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of BRCA2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic. -

The c.6676_6677delGA pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a deletion of two nucleotides at nucleotide positions 6676 to 6677, causing a translational frameshift with a predicted alternate stop codon (p.E2226Sfs*6). This alteration was identified in 1/527 index patients with either breast, ovarian, or pancreatic cancer. (Loizidou MA et al. Clin. Genet. 2017 Apr;91:611-615) as well as a male breast cancer patient (Ibrahim M et al. BMC Cancer 2018 02;18:179). This alteration was also identified in a large, worldwide study of BRCA1/2 mutation positive families (Rebbeck TR et al. Hum. Mutat. 2018 05;39:593-620). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
