This variant has been reported in at least two individuals affected with breast or ovarian cancer (PMID: 12655560, 24156927, 26843898) and has been identified in 2 families among the CIMBA participants (PMID: 29446198). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of BRCA2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic. -
The c.9027delT pathogenic mutation, located in coding exon 22 of the BRCA2 gene, results from a deletion of one nucleotide at nucleotide position 9027, causing a translational frameshift with a predicted alternate stop codon (p.H3010Ifs*18). This mutation has been reported in multiple individuals with Hereditary Breast and Ovarian Cancer (HBOC) syndrome (Manguoglu AE et al. Hum. Mutat. 2003 Apr; 21(4):444-5; Tea MK et al. Maturitas 2014 Jan; 77(1):68-72; Wojcik P et al. Hered Cancer Clin Pract 2016;14:5; Rebbeck TR et al. Hum. Mutat., 2018 05;39:593-620). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -

Truncations downstream of this position have been classified as pathogenic by our laboratory (e.g. c.9076C>T (p.Gln3026X), c.9093_9094delinsG (p.Thr3033fsX29), c.9097dupA (p.Thr3033fsX11)). The variant has been reported in multiple HBOC affected individuals in literature (Manguoglu 2003, Tea 2014, Wojcik 2016). This variant is absent in 245484 control chromosomes. In addition, multiple clinical diagnostic laboratories/reputable databases classified this variant as pathogenic. Taken together, this variant is classified as pathogenic. -
