Genes affected
BRCA2 (HGNC:1101): (BRCA2 DNA repair associated) Inherited mutations in BRCA1 and this gene, BRCA2, confer increased lifetime risk of developing breast or ovarian cancer. Both BRCA1 and BRCA2 are involved in maintenance of genome stability, specifically the homologous recombination pathway for double-strand DNA repair. The largest exon in both genes is exon 11, which harbors the most important and frequent mutations in breast cancer patients. The BRCA2 gene was found on chromosome 13q12.3 in human. The BRCA2 protein contains several copies of a 70 aa motif called the BRC motif, and these motifs mediate binding to the RAD51 recombinase which functions in DNA repair. BRCA2 is considered a tumor suppressor gene, as tumors with BRCA2 mutations generally exhibit loss of heterozygosity (LOH) of the wild-type allele. [provided by RefSeq, May 2020]

Aberrant splicing and/or loss of function is an established mechanism of disease. This prediction has been confirmed by an RNA splicing study (PMID: 29881398).This variant has been reported in multiple individuals with hereditary breast and ovarian cancer syndrome (PMID: 23593081, 30078507, 31825140, 29446198). Pathogenic, criteria provided, single submitterclinical testingAmbry GeneticsAug 07, 2018The c.7805+1G>A intronic pathogenic mutation results from a G to A substitution one nucleotide after coding exon 15 of the BRCA2 gene. A minigene assay demonstrated that this alteration results in total or partial skipping of coding exon 15 in over 90% of transcripts (Fraile-Bethencourt E et al. Front Genet. 2018 May;9:188). It was also identified in a patient with pancreatic cancer (Shindo K et al. J. Clin. Oncol. 2017 Oct;35(30):3382-3390). In addition to the clinical data presented in the literature, alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. As such, this alteration is classified as a disease-causing mutation. -
Pathogenic, criteria provided, single submitterclinical testingColor Diagnostics, LLC DBA Color HealthMar 09, 2022This variant causes a G to A nucleotide substitution at the +1 position of intron 16 of the BRCA2 gene. RNA studies have shown that this variant causes out-of-frame skipping of exon 16, predicted to result in a frameshift and premature translation stop signal (PMID: 29881398). This variant has been reported in individuals affected with pancreatic cancer, early-onset breast cancer (PMID: 23593081, 28767289) and has been identified in 2 families among the CIMBA participants (PMID: 29446198). This variant has been identified in 1/251220 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Loss of BRCA2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic. -
Hereditary breast ovarian cancer syndrome Pathogenic:2
Pathogenic, criteria provided, single submitterclinical testingInvitaeJul 02, 2023For these reasons, this variant has been classified as Pathogenic. Studies have shown that disruption of this splice site alters mRNA splicing and is expected to lead to the loss of protein expression (PMID: 29881398). ClinVar contains an entry for this variant (Variation ID: 52410). Disruption of this splice site has been observed in individual(s) with pancreatic cancer and/or ovarian cancer (PMID: 28767289, 30078507). This variant is present in population databases (rs81002809, gnomAD 0.0009%). This sequence change affects a donor splice site in intron 16 of the BRCA2 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584). -
Pathogenic, criteria provided, single submitterclinical testingGeneKor MSAJan 01, 2020This variant is a substitution of the first nucleotide base of intron 16 of BRCA2 gene and is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product. -
