FH (HGNC:3700): (fumarate hydratase) The protein encoded by this gene is an enzymatic component of the tricarboxylic acid (TCA) cycle, or Krebs cycle, and catalyzes the formation of L-malate from fumarate. It exists in both a cytosolic form and an N-terminal extended form, differing only in the translation start site used. The N-terminal extended form is targeted to the mitochondrion, where the removal of the extension generates the same form as in the cytoplasm. It is similar to some thermostable class II fumarases and functions as a homotetramer. Mutations in this gene can cause fumarase deficiency and lead to progressive encephalopathy. The substitution creates a nonsense variant, which changes a Glutamic Acid to a premature stop codon (GAA>TAA), and is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. This variant, also published as Glu270Ter using alternate nomenclature, has been reported in at least two individuals with a personal/family history of multiple cutaneous leiomyomas (Gardie 2011). We consider FH Glu313Ter to be pathogenic. -

This changes the amino acid from a glutamic acid to a stop codon within coding exon 7. This mutation has been reported in two French families with hereditary leiomyomatosis and renal cell cancer (HLRCC) (Gardie B et al. J. Med. Genet. 2011 Apr;48(4):226-34). Of note, this alteration is also designated as 808G>T and Glu270X in published literature. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
