Gorlin syndrome Pathogenic:1
This sequence change creates a premature translational stop signal (p.Trp948Argfs*8) in the PTCH1 gene. It is expected to result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with PTCH1-related conditions. Loss-of-function variants in PTCH1 are known to be pathogenic (PMID: 16301862, 16419085). For these reasons, this variant has been classified as Pathogenic. -
Hereditary cancer-predisposing syndrome Pathogenic:1
The c.2842_2849delTGGGTCCA pathogenic mutation, located in coding exon 17 of the PTCH1 gene, results from a deletion of 8 nucleotides at nucleotide positions 2842 to 2849, causing a translational frameshift with a predicted alternate stop codon (p.W948Rfs*8). This alteration has been observed in at least one individual with a personal and/or family history that is consistent with PTCH1-related disease (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
