The p.K433* pathogenic mutation (also known as c.1297A>T), located in coding exon 11 of the PMS2 gene, results from an A to T substitution at nucleotide position 1297. This changes the amino acid from a lysine to a stop codon within coding exon 11. This alteration has been reported as pathogenic in 1/1197 individuals from Greece, Romania, and Turkey undergoing evaluation for inherited cancer predisposition (Tsaousis GN et al. BMC Cancer, 2019 Jun;19:535). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
This variant is a single amino acid change from Lysine to a premature translational stop signal at codon 433 of the PMS2 protein. This is expected to result in a truncated, non-functional protein product. Truncating variants in the PMS2 are known to be pathogenic (PMID: 21376568, 24362816).
