Pathogenic, criteria provided, single submitter | clinical testing | GeneDx | Dec 24, 2018 | This deletion of one nucleotide in BRIP1 is denoted c.1236delA at the cDNA level and p.Val413PhefsX10 (V413FfsX10) at the protein level. The normal sequence, with the base that is deleted in brackets, is CAGA[delA]GTTC. The deletion causes a frameshift, which changes a Valine to a Phenylalanine at codon 413 and creates a premature stop codon at position 10 of the new reading frame. This variant is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. BRIP1 c.1236delA has been reported in at least two individuals with ovarian cancer (Norquist 2015, Ramus 2015). We consider this variant to be pathogenic. - |

Pathogenic, criteria provided, single submitter | clinical testing | Quest Diagnostics Nichols Institute San Juan Capistrano | Oct 17, 2018 | The variant results in a shift of the reading frame, and is therefore predicted to significantly disrupt the protein structure. Found in at least one symptomatic patient, and found in general population data that is consistent with pathogenicity. - |

Pathogenic, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Jan 09, 2024 | This sequence change creates a premature translational stop signal (p.Val413Phefs*10) in the BRIP1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in BRIP1 are known to be pathogenic (PMID: 16116423, 17033622, 21964575). This variant is present in population databases (no rsID available, gnomAD 0.002%). This premature translational stop signal has been observed in individual(s) with ovarian cancer (PMID: 26315354). ClinVar contains an entry for this variant (Variation ID: 216128). For these reasons, this variant has been classified as Pathogenic. - |
Hereditary cancer-predisposing syndrome Pathogenic:2
Pathogenic, criteria provided, single submitter | clinical testing | Ambry Genetics | Apr 26, 2022 | The c.1236delA pathogenic mutation, located in coding exon 8 of the BRIP1 gene, results from a deletion of one nucleotide at nucleotide position 1236, causing a translational frameshift with a predicted alternate stop codon (p.V413Ffs*10). This mutation has been identified in multiple patients with ovarian cancer (Ramus SJ et al. J. Natl. Cancer Inst. 2015 Nov;107; (Norquist BM et al. JAMA Oncol. 2016 Apr;2:482-90). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. - |
Pathogenic, criteria provided, single submitter | clinical testing | Color Diagnostics, LLC DBA Color Health | Sep 19, 2022 | This variant deletes 1 nucleotide in exon 9 of the BRIP1 gene, creating a frameshift and premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. This variant has been reported in two individuals affected with ovarian cancer (PMID: 26315354, 26720728). This variant has been identified in 2/282514 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Loss of BRIP1 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic. - |

Pathogenic, criteria provided, single submitter | clinical testing | Myriad Genetics, Inc. | Jun 01, 2023 | This variant is considered pathogenic. This variant creates a frameshift predicted to result in premature protein truncation. - |
