The normal sequence, with the base that is deleted in brackets, is GCTA[delT]ATCA. The deletion causes a frameshift which changes an Isoleucine to an Asparagine at codon 766, and creates a premature stop codon at position 46 of the new reading frame. This variant is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. MSH2 c.2297delT has been observed in at least one individual with early onset colon cancer, who met Amsterdam II criteria (Limburg 2011). We consider this variant to be pathogenic. -

To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar and all laboratories classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -
