Genes affected
JPH2 (HGNC:14202): (junctophilin 2) Junctional complexes between the plasma membrane and endoplasmic/sarcoplasmic reticulum are a common feature of all excitable cell types and mediate cross talk between cell surface and intracellular ion channels. The protein encoded by this gene is a component of junctional complexes and is composed of a C-terminal hydrophobic segment spanning the endoplasmic/sarcoplasmic reticulum membrane and a remaining cytoplasmic domain that shows specific affinity for the plasma membrane. This gene is a member of the junctophilin gene family. Alternative splicing has been observed at this locus and two variants encoding distinct isoforms are described. [provided by RefSeq, Jul 2008]

The p.Ser171_Asn172dup variant in JPH2 has not been previously reported in indiv iduals with cardiomyopathy. Data from large population studies is insufficient t o assess the frequency of this variant. This variant results in a duplication of 2 amino acids at position 172. It is unclear if this duplication will impact th e protein. In summary, the clinical significance of the p.Ser171_Asn172dup varia nt is uncertain. -

This variant, c.511_516dup, results in the insertion of 2 amino acid(s) of the JPH2 protein (p.Ser171_Asn172dup), but otherwise preserves the integrity of the reading frame. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with JPH2-related conditions. ClinVar contains an entry for this variant (Variation ID: 218769). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
