The c.2155_2157dupCTA variant (also known as p.L719dup), located in coding exon 14 of the NBN gene, results from an in-frame duplication of CTA at nucleotide positions 2155 to 2157. This results in the duplication of an extra leucine residue between codons 719 and 720. This amino acid position is highly conserved in available vertebrate species. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
